Automated Analysis & Sorting of Drosophila' Imaginal Discs  
Drosophila Fluorescent-Imaginal Disc Analysis and Sorting using the COPAS™ Flow Cytometry Technique.

The COPAS SELECT instrument from Union Biometrica automates the analysis and sorting of Drosophila embryos and larvae on the basis of object size and fluorescence signals. In addition to sorting whole organisms, another area of interest has been to use the instrument for the separation of fluorescent tissues from Drosophila larvae.


A poster was presented at the recent 45th Annual Drosophila Research Conference in which we tested two protocols for disrupting larvae for the extraction of intact imaginal discs. We then demonstrated the ability to isolate imaginal discs from a crude tissue sample from Drosophila larvae using the COPAS instrument.

About the instrument: Based on similar principles as flow cytometry, the COPAS™ instrument from Union Biometrica provides an automated alternative to manual sorting for increased speed, sensitivity, quantification, and repeatability of experiments. The COPAS SELECT instrument analyzes and sorts small model organisms and other objects (from 120-300 microns) on the basis of size, density, and fluorescence signals. A gentle pneumatic sorting mechanism is used to permit the analysis and handling of sensitive organisms and tissues with a high level of recovery and a low level of lethality.
